There are two variants of CJC-1295:
The first variant, GHRH, is produced in the hypothalamus and releases growth hormone in a pulsatile fashion from the pituitary. However, it has a very short half-life of just a few minutes, leading to rapid elimination from the body.
The active segment of GHRH consists of the initial 29 amino acids and is available commercially as Sermorelin, which has been modified to extend its half-life to approximately 30 minutes. This is known as CJC-1295. Further modifications, including the addition of DAC (Drug Affinity Complex), prolong its half-life to around 8 days, resulting in the variant CJC-1295 + DAC. Alternatively, CJC-1295 can be formulated in a non-DAC form, which creates a more natural growth hormone spike each night.
This extended half-life allows for just one or two injections per week; however, the sustained levels may provide a continuous stimulus for GH release from the pituitary via the GHRH receptor, which may not be physiological. This could potentially reduce the amplitude of GH pulses and subsequently decrease stimulation of GH-responsive tissues.
Safety Considerations:
It is recommended to take 'hormone holidays' every three to six months while using long-acting CJC forms, allowing the pituitary to recover. During these breaks, Sermorelin can be used as an alternative to CJC-1295 + DAC.
Implementing these 'hormone holidays' may also help mitigate the risk of developing GH resistance, which could arise from the formation of antibodies that bind to and deactivate GH or a reduction of GH receptors on tissues (down-regulation). While these concerns are theoretical, long-term studies have not confirmed them.
Possible side effects of CJC-1295 may include injection site reactions (irritation, redness, swelling, pain, itching), headaches, diarrhea, flushing (vasodilation), temporary low blood pressure, nausea, and abdominal discomfort.
