GHRH is naturally synthesized in the hypothalamus and stimulates the pituitary gland to release GH in a pulsatile manner. However, GHRH has a very short half-life of just a few minutes, leading to rapid elimination from the bloodstream and brief effects.
Types of CJC-1295: The active part of GHRH consists of the first 29 amino acids, available commercially as Sermorelin. This was further optimized to extend its half-life to 30 minutes, resulting in CJC-1295. Enhanced with a Drug Affinity Complex (DAC), CJC-1295 has a prolonged half-life of approximately 8 days, known as CJC-1295 + DAC. A non-DAC version is also available that mimics the natural nightly GH spike.
The extended half-life of CJC-1295 + DAC allows for injections just once or twice a week. However, this extended presence can lead to constant stimulation of GH release that may not mimic natural physiological patterns, potentially reducing GH pulse amplitude and leading to diminished effects on GH-responsive tissues.
For optimal use of long-acting CJC molecules, it is recommended to take "hormone holidays" every three to six months for three months at a time, allowing the pituitary gland to recover. During these breaks, Sermorelin serves as an ideal substitute for CJC-1295 + DAC.
Safety Considerations: These hormone holidays may also mitigate the risk of developing GH resistance, which can occur from the formation of antibodies against GH or a decrease in GH receptors on tissues (down-regulation).
